Protein corona modulates interaction of spiky nanoparticles with lipid bilayers

چکیده
The impact of protein corona on the interactions nanoparticles (NPs) with cells remains an open question. This question is particularly relevant to NPs which sizes, ranging from tens hundreds nanometers, are comparable sizes most abundant proteins in plasma. Protein match typical thickness various coatings and ligands layers, usually present at surfaces larger NPs. Such size may affect properties designed function We offer a direct demonstration how can dramatically change interaction mode between lipid bilayers. To this end, we choose extreme case NP surface modification: nanostructures form rigid spikes 10–20 nm length gold nanoparticles. In absence observe formation reversible pores when spiky adsorb contrast, presence bovine serum albumin (BSA) adsorbed spiked NPs, effectively reduces exposed Thus, changes qualitatively dynamics pore formation, completely suppressed high concentrations. These results suggest that not only be critical for membranes, it their interaction, thus offsetting role chemistry ligands.

متن کاملDNA interaction with freestanding cationic lipid bilayers
We study the interaction of ds-DNA molecules with freestanding and supported cationic lipid bilayers. DNA fragments of different lengths (5, 10, 20, and 48 kbp) were adsorbed on lipid membranes consiting mainly of zwitterionic DOPC with added fractions of cationic DOTAP (1...10 %). Freestanding bilayers were modeled using giant unilamellar vesicles of sizes >100 μm. Supported lipid bilayers wer...
متن کاملInteraction of Daptomycin with Lipid Bilayers: A Lipid Extracting Effect
Daptomycin is the first approved member of a new structural class of antibiotics, the cyclic lipopeptides. The peptide interacts with the lipid matrix of cell membranes, inducing permeability of the membrane to ions, but its molecular mechanism has been a puzzle. Unlike the ubiquitous membrane-acting host-defense antimicrobial peptides, daptomycin does not induce pores in the cell membranes. Th...
متن کاملPhase Behavior and Structure Properties of Supported Lipid Monolayers and Bilayers in Interaction with Silica Nanoparticles
In this study we investigate silica nanoparticle (SiO2NP) effects on the structure and phase properties of supported lipid monolayers and bilayers, coupling surface pressure measurements, fluorescence microscopy and atomic force microscopy. SiO2-NPs typically in size range of 10nm to 100 nm in diameter are tested. Our results suggest first that lipid molecules organization depends to their natu...
